Sequenom’s automated DNA analysis platform has recently been broadly used in a variety of species important in agriculture and livestock. A single MassARRAY system can be used for the analysis of DNA sequence variations like SNPs, gene expression, DNA methylation, copy number variation and certain DNA sequencing applications. The sensitivity is 100 to 1,000 times higher than real time PCR and the quantification is an order of magnitude better than real time PCR. But perhaps the strongest feature of the technology is the ease and flexibility with which new problem-specific applications can be developed and validated. This is especially important for agribiotech where so many diverse species are of interest. For example, with commercial livestock and crops, panels of 10s to 100s of SNPs can be rapidly developed that are extremely powerful for individual identification, breeding, and feedlot/nutrient management. The increased use of high content discovery technologies such as 2nd Generation Sequencing and microarrays in agricultural research has resulted in a clear need for validation of discovered markers. With plants, ploidies higher than 2, mean that essentially every locus must be treated as a potential copy number variant, and mass spectrometry offers a facile way to quantify such variants in a cost-effective multiplexed manner. DNA mass spectrometry also shows great promise in the automated identification and typing of micro-organisms, and thus might be very useful in the study of a wide variety of plant and animal infectious diseases. This seminar will review the basics of the technology and then illustrate a number of current case studies of the above applications.
